For anyone implemented this idea will appreciate for the how to guide.

1) One postfix server in DMZ with a different server as the content filter for checking virus and spam
2) Another postfix server in LAN with a different  dovecot server and the mail folders
3) Cluster for Postfix/Dovecot server for HA and Load Balance.

1-2) Better to place Antispam/Antivirus gateway and Mail server together in DMZ, you can make a 2 servers - one will be real one and another can be MX backup server.
Another scenario: Make MX backup server like Antispam/Antivirus gateway in DMZ just to relaying all emails to the real second Mail server in LAN
