I have only a Global Spam Policy defined on the server, nothing for domains and users.

If I want to add whitelists and blacklists for a domain or user, should I create also first create a spam policy for that domain or user for the blacklists and whitelists to be effective?

No. Spam policy and white/blacklists are separated.
