I tried to open port 465 by following iredmail documentation, https://docs.iredmail.org/enable.smtps.html

465 is used for SMTP over SSL and is deprecated.
If you want to test it, you need to choose port 25 as it is used by your mailserver to recive mail from foreign mail servers, then it should deliver.

I have LetsEncrypt ssl, I want to use port 465 over SSL

Why you want to use SSL?
SSL can't be used, even if you want to send mails trough port 465, it needs to be TLS.

Like i said, port 25 is used by your mailserver to recive mail from foreign mail servers, 465 is SMTPS, which is depecated, should not be used at all, and does not work without authentication

Port 465 is smtp over ssl, although it's deprecated but still many companies use it. I suggest you use port 587 (smtp over tls) instead.
