That's blacklist. do you have any issue with white/blacklist managed with iRedAdmin-Pro?

We have done as like the instruction of that link but spam message does not send to junk.

Can you show headers of an example of a spam e-mail?

Please turn on debug mode in Dovecot to see why it doesn't execute this global sieve script. Reference:
http://www.iredmail.org/docs/debug.dovecot.html

Also, per-domain or per-user transport must be 'dovecot', so that received mails will be delivered locally with Dovecot LDA with sieve support, otherwise it won't trigger sieve at all.
